No. Optimization always works in the same manner regardless of load algorithm that you select in Preferences > Algorithm Settings > Algorithm. 

Optimization occurs after the route is built. DirectRoute performs additional passes over the routing data, while considering penalty factors, time windows, work time, and more, to look for options that would allow stops to be moved either within each route, or between routes, in an effort to return a lower-cost routing solution.
